[Abstract]

Objectives: To investigate p53 gene mutations of colorectal cancer in Guangxi and their relation to clinicopathological features for later gene diagnosis and gene therapy. Method: Mutations were screened with polymerase chain reaction-single strand conformation polymorphisms (PCR-SSCP) technique .The p53 gene were analysed in 45 cases of colorectal cancer (90 chromosomes in all). Results: Mutations of p53 gene were found in 33.3% colorectal cancers and a case of compound-heterozygote mutation was found. Mutations of p53 gene lay in codon 175(CGC?CAC)?codons 177?182?-CCCCACCATGAGCGCTGC??codon 124?TGC?GGC??codon 248?CGG?CAG??codon 257?-TG??codon 246?ATG?GTG??codon 258?GAA?TAA??codon 272?GTG?ATG??IVS-?-1?G?A?,IVS-?-1?G?C?and Codon 317(-C). Codon 175(CGC?CAC) taked the most among all mutations ,which accounted for 31.6% .The<WP=6>second was codons 177?182?-CCCCACCATGAGCGCTGC?, which accounted for 12.5%. Each of other mutations accounted for 6.25%. Deletion mutation of codons 177?182?-CCCCAC CATGAGCGCTGC? and missence mutation of codon 124 ?T?A>G:C?hadn?t been found in IARC Tp53 mutation database.Conclusions: Missence mutation and somatic mutation occupied the most among all mutations of colorectal cancer in Guangxi. Mutation of p53 gene were associated with infiltrating depth and differentiation of colorectal cancer (P< 0.05). PCR-SSCP technique was a simple and effective method for gene diagnosis .
